Felix Baumgartner?s record-breaking jump captivates Internet, Twitter

Felix Baumgartner's record-setting jump from the edge of space captivated millions watching the Red Bull-sponsored event live online via YouTube. And the 43-year-old Austrian skydiver's two-hour, 24-plus-mile balloon ascent and harrowing four-minute freefall in a pressurized spacesuit provided plenty of fodder for those on Twitter.

"And you thought your weekend was exciting," Twitter user @wonderwamy wrote.

"I seem to have tuned into the part of #spacejump where ground control is explaining to Baumgartner how to build an IKEA bookshelf," Dave Itzkoff wrote on Twitter.

"Instagram it or it didn't happen," Kathleen Schmidt tweeted.

The Red Bull tie-in, though, was mocked by many.

"My only question is: How will 5-Hour Energy top this?" Lindsey Weber asked.

Several Twitter users thanked Baumgartner in advance for a Halloween costume idea. And at least one wondered where the audience went for his jump. "Guys, I'm up next," the user @Scharpling wrote. "Where is everybody?"

Baumgartner's soft landing in the New Mexico desert was applauded, too.

"Best landing since McKayla Maroney," Dave Bry wrote, a nod to the gold-medal-winning American gymnast.

"I can't land like that jumping off a stepladder," Steve Yelvington tweeted.

At its peak, more than 8 million users watched Baumgartner's freefall, according to YouTube.

"Not confirmed yet, but I'm hearing this was the most live-tweeted space jump in history," Joseph Weisenthal joked.

"So there's your new [business] model, Web guys," Peter Kafka wrote. "Just gotta launch a guy into space multiple times a day."

"Red Bull wins the internet for today," @JMRooker added. "Everybody can go home now."

Complex logic circuit made from bacterial genes

ScienceDaily (Oct. 12, 2012) ? By force of habit we tend to assume computers are made of silicon, but there is actually no necessary connection between the machine and the material. All that an engineer needs to do to make a computer is to find a way to build logic gates -- the elementary building blocks of digital computers -- in whatever material is handy.

So logic gates could theoretically be made of pipes of water, channels for billiard balls or even mazes for soldier crabs.

By comparison Tae Seok Moon's ambition, which is to build logic gates out of genes, seems eminently practical. As a postdoctoral fellow in the lab of Christopher Voigt, PhD, a synthetic biologist at the Massachusetts Institute of Technology, he recently made the largest gene (or genetic) circuit yet reported.

Moon, PhD, now an assistant professor of energy, environmental and chemical engineering in the School of Engineering & Applied Science at Washington University in St. Louis is the lead author of an article describing the project in the Oct. 7 issue of Nature. Voigt is the senior author.

The tiny circuits constructed from these gene gates and others like them may one day be components of engineered cells that will monitor and respond to their environments.

The number of tasks they could undertake is limited only by evolution and human ingenuity. Janitor bacteria might clean up pollutants, chemical-engineer bacteria pump out biofuels and miniature infection-control bacteria might bustle about killing pathogens.

How to make an AND gate out of genes

The basis of modern computers is the logic gate, a device that makes simple comparisons between the bits, the 1s and 0s, in which computers encode information. Each logic gate has multiple inputs and one output. The output of the gate depends on the inputs and the operation the gate performs.

An AND gate, for example, turns on only if all of its inputs are on. An OR gate turns on if any of its inputs are on.

Suggestively, genes are turned on or off when a transcription factor binds to a region of DNA adjacent to the gene called a promotor.

To make an AND gate out of genes, however, Moon had to find a gene whose activation is controlled by at least two molecules, not one. So only if both molecule 1 AND molecule 2 are present will the gene be turned on and translated into protein.

Such a genetic circuit had been identified in Salmonella typhimurium, the bacterium that causes food poisoning. In this circuit, the transcription factor can bind to the promotor of a gene only if a molecule called a chaperone is present. This meant the genetic circuit could form the basis of a two-input AND gate.

The circuit Moon eventually built consisted of four sensors for four different molecules that fed into three two-input AND gates. If all four molecules were present, all three AND gates turned on and the last one produced a reporter protein that fluoresced red, so that the operation of the circuit could be easily monitored.

In the future, Moon says, a synthetic bacterium with this circuit might sense four different cancer indicators and, in the presence of all four, release a tumor-killing factor.

Crosstalk and timing faults

There are huge differences, of course, between the floppy molecules that embody biological logic gates and the diodes and transistors that embody electronic ones.

Engineers designing biological circuits worry a great deal about crosstalk, or interference. If a circuit is to work properly, the molecules that make up one gate cannot bind to molecules that are part of another gate.

This is much more of a problem in a biological circuit than in an electronic circuit because the interior of a cell is a kind of soup where molecules mingle freely.

To ensure that there wouldn't be crosstalk among his AND gates, Moon mined parts for his gates from three different strains of bacteria: Shigella flexneri and Pseudomonas aeruginosa, as well as Salmonella.

Although the parts from the three different strains were already quite dissimilar, he made them even more so by subjecting them to error-prone copying cycles and screening the copies for ones that were even less prone to crosstalk (but still functional).

Another problem Moon faced is that biological circuits, unlike electronic ones, don't have internal clocks that keep the bits moving through the logic gates in lockstep. If signals progress through layers of gates at different speeds, the output of the entire circuit may be wrong, a problem called a timing fault.

Experiments designed to detect such faults in the synthetic circuit showed that they didn't occur, probably because the chaperones for one layer of logic gates degrades before the transcription factors for the next layer are generated, and this forces a kind of rhythm on the circuit.

Hijacking a bacterium's controller

"We're not trying to build a computer out of biological logic gates," Moon says. "You can't build a computer this way. Instead we're trying to make controllers that will allow us to access all the things biological organisms do in simple, programmable ways."

"I see the cell as a system that consists of a sensor, a controller (the logic circuit), and an actuator," he says. "This paper covers work on the controller, but eventually the controller's output will drive an actuator, something that will do work on the cell's surroundings. "

An synthetic bacterium designed by a friend of Moon's at Nanyang Technological University in Singapore senses signaling molecules released by the pathogen Pseudomonas aeruginosa. When the molecules reach a high enough concentration, the bacterium generates a toxin and a protein that causes it to burst, releasing the toxin, and killing nearby P. aeruginosa.

"Silicon cannot do that," Moon says.

New rules, tough talk as Singapore seeks to end tax haven image

SINGAPORE (Reuters) - Billionaire Australians buying exclusive condominiums. Germans moving money from Swiss accounts. Secretive banking laws. Asia's premier wealth management centre. Low tax rates.

As Singapore revels in its reputation as an open economy with the world's highest concentration of millionaires, the tiny island of 5.3 million people is also accused of being a magnet for tax evaders - an image it is vehemently seeking to banish.

Amid German concerns that its wealthy citizens are moving funds from Switzerland before a new German-Swiss tax treaty takes effect next year, Singapore and Germany said on Sunday they had agreed to bolster their double-taxation agreement with internationally agreed standards on information sharing.

"Banking secrecy will not constitute an obstacle to exchanging information," said the joint statement, which came at the end of German Finance Minister Wolfgang Schaeuble's weekend visit to Singapore.

Media reports have put the amount of German money moving to Singapore in the double-digit billions.

"The perception is that Swiss banks have concluded Switzerland is unlikely to remain a tax haven for much longer and Singapore is the new place to do business," said Ronen Palan, a professor at City University London who has conducted numerous studies on offshore finance.

Swiss banks could see assets from Western European clients fall 28 percent to 623 billion Swiss francs ($668 billion) by 2014 because of the deals to tax undeclared accounts, the Boston Consulting Group said in a report in May.

Singapore and its rival Hong Kong look set to benefit.

Together, the two Asian hubs manage $1 trillion in offshore funds, with about 75 percent of that coming from within the region. But Singapore and Hong Kong may overtake Switzerland - now the largest offshore wealth centre with assets of about $2.1 trillion - in 15 to 20 years, Boston Consulting said.

"INTEGRITY AND REPUTATION"

Singapore, with tax rates that top out at 20 percent and no capital gains tax, is already synonymous with wealth. BMW and Mercedes were the top two brands among all cars sold in the first eight months of this year, bestsellingcarsblog.com says.

Safe and clean, the city-state bills itself as a tropical refuge with exclusive residential enclaves, a marina for super-yachts, two casinos, fine dining, high-end boutiques and an annual Formula One race that brings in the global jet-set.

Rich residents include Eduardo Saverin, the co-founder of Facebook, who has called Singapore home since 2009.

Brazilian-born Saverin, who renounced his U.S. citizenship this year, was in eighth spot on a Singapore rich list published by Forbes Magazine with an estimated net worth of $2.2 billion.

Locals who made fortunes in real estate, finance and trading figured prominently but the list also included New Zealand-born investor Richard Chandler with $2.9 billion and China-born property developer Zhong Sheng Jian with $1.4 billion.

A 10 percent property duty imposed on foreigners, part of efforts to cool the housing market, has done little to dissuade the ultra-wealthy - many of them Chinese, Indian, Malaysian and Indonesian - from ploughing money into Singapore real estate.

Australian mining tycoons are also moving in. Gina Rinehart reportedly paid S$57 million ($46.7 million) for two units at Seven Palms Sentosa Cove, a luxury beachfront condominium, while Nathan Tinkler recently moved his family to Singapore.

But authorities bristle at any suggestion of tax dodging.

A short paragraph on page 17 of an Indian government white paper on "black money" was inflammatory enough to prompt Singapore's prime minister, Lee Hsien Loong, to make a formal complaint in July to his Indian counterpart Manmohan Singh.

The paper did not directly call Singapore a tax haven but suggested figures showing it accounts for nearly 10 percent of foreign direct investment in India could be due to Indian nationals routing money through the city-state to avoid taxes.

"We have demarched the Indian government on this matter to put this record straight and explain why this is not true and it has been mistaken," Lee said during a visit to New Delhi.

Over the past three years, Singapore has upgraded half of its 70 tax treaties with other countries to make it easier to exchange information on possible tax dodgers.

From next year, bankers who help clients evade tax risk ending up in court on money laundering charges.

The new rules are part of "efforts to protect the integrity and reputation of Singapore as a trusted international financial centre," the Monetary Authority of Singapore (MAS), the central bank and financial regulator, said last week.

The first stage of a peer review by the Organisation for Economic Co-operation and Development in 2011 found Singapore had most of the agreed standards in place but needed to continue to update and expand its information exchange pacts.

Thomas Eigenthaler, chairman of the German Tax Union, welcomed an upgrade to Germany and Singapore's "rudimentary, fragmentary" tax agreement but said "the decisive thing will be how it is actually implemented."

BENEFITS OF TRANSPARENCY

Palan at City University London said trying to assess the city-state's credentials is tricky, partly due to difficulties getting detailed information about its offshore finance sector.

"We cannot get any access to Singapore and that for us does raise flags," he said. "It's easier to do research on Jersey or Switzerland but in Singapore it's almost impossible."

Before the start of the global financial crisis in 2008, most questions about Singapore's offshore role focused on the billions of dollars kept in the city-state by rich Indonesians.

Now, as cash-strapped Western governments ramp up their efforts to improve tax collection and Swiss banks are forced to open up their books, Singapore is facing renewed accusations that some of the funds flowing in may be illicit.

Lawyers say much of the recent inflows reflect the growing number of Asian millionaires, the region's strong growth and the fact that some legitimate funds are choosing to abandon offshore centers with less savory reputations in favor of Singapore.

As more tax-paying millionaires want to get exposure to Asian growth, they say, Singapore must do all it can to show it will not tolerate black money.

"Singapore stands to gain a lot from proactively embracing the move towards global tax transparency," said Dawn Quek, a senior associate at law firm Baker & McKenzie, Wong & Leow.

The MAS is pushing banks to toughen scrutiny of clients with plans to designate a variety of serious tax crimes as predicates for money laundering offences from July 2013.

In 2010 and 2011, a total of 44 people were convicted of money laundering offences in Singapore, mostly related to fraud, and close to S$130 million ($106.5 million) was seized or frozen, the Commercial Affairs Department said.

"Singapore has always taken a tough enforcement approach against money laundering and our authorities will not hesitate to pursue and prosecute such cases," MAS told Reuters.
